Il film inizia nel XVI secolo (1519) a Seville (nel sud della Spagna) e narra di due uomini, Tulio e Miguel. Durante una partita a dadi, usando dadi truccati, vincono una mappa che indicherebbe la locazione della leggendaria città dell'oro nel Nuovo Mondo, El Dorado. Viene però scoperto il loro trucco al gioco e come risultato i due sono costretti a scappare, finendo per sbaglio con l'imbarcarsi clandestinamente sulla flotta di Hernán Cortés diretta verso il Mexico. Vengono scoperti e imprigionati ma riescono scappare con una scialuppa con l'intelligente cavallo di Cortés e successivamente scoprono la città nascosta di El Dorado, dove vengono accolti e venerati come dei. I due però dovranno provare i loro poteri divini al sanguinario sacerdote Tzekel-Kan. In loro aiuto verrà Chel, astuta e affascinante ragazza indigena
