A personagem principal é o Dr. Gregory House, interpretado pelo actor britânico Hugh Laurie.
House (uma espécie de Sherlock Holmes da medicina) é um infectologista e nefrologista que se destaca não só pela capacidade de elaborar diagnósticos diferenciais, como também pelo seu mau-humor, cepticismo, narcisismo e pelo seu distanciamento dos pacientes, já que ele considera completamente desnecessário interagir com eles.
A acção passa-se num lugar ficcional chamado Hospital de Princeton-Plainsboro, em Princeton.
Quatro temporadas da série já foram produzidas até ao momento. É exibida no Brasil pelo Universal Channel e pela Rede Record e em Portugal pela TVI e pela Fox.
