Il film inizia con l'uccisione di tre killer presso una stazione ferroviaria da parte di un misterioso straniero (Charles Bronson), soprannominato "Armonica" per l'abitudine di vagare suonando continuamente questo strumento musicale.


I tre sono uomini di Frank (Henry Fonda), spietato assassino al soldo di Morton (Gabriele Ferzetti). Costui è un moribondo industriale ferroviario ammalato di tubercolosi ossea che vive e dirige i propri affari sul suo treno personale, smanioso di terminare, prima di morire, l'ultimo tratto di binari che unisce la costa orientale degli U.S.A. a quella occidentale. Nel duello Armonica rimane ferito e si rifugia in una squallida locanda. Qui trova Jill (Claudia Cardinale), una bellissima donna proveniente da New Orleans non proprio a suo agio nel selvaggio West, e Cheyenne (Jason Robards), pericoloso bandito appena evaso dal carcere........
