La trama del film ricalca più o meno fedelmente la trama del romanzo. Nel 1327, alcuni terribili omicidi sconvolgono un'abbazia benedettina sperduta sui monti del Nord-Italia. Nel monastero dovrà svolgersi un importante concilio francescano a cui è chiamato a partecipare il dotto frate Guglielmo da Baskerville (Sean Connery). Nel contempo, l'abate affida a Guglielmo le indagini degli omicidi in virtù della sua esperienza di inquisitore, senza dimenticare le vociferazioni sull'Anticristo che da sempre circolano nell'abbazia. Il francescano, insieme al suo giovane novizio Adso, si ritrova in un ambiente ostile, un'abbazia piena di libri e di cultura ma anche segreta e spaventosa, su cui dovrà indagare prima dell'arrivo della Santa Inquisizione.
