Fred, Velma, Daphne und Shaggy sind vier Schüler an der Coolsville High. Als es in der Schule zu mysteriösen Ereignissen kommt, bei denen auch die Fassade des Gebäudes verunstaltet wird, geraten die vier in Verdacht, eine Geistererscheinung inszeniert zu haben. Vom Schulleiter umgehend suspendiert, schließen sie sich zur "Monster AG" zusammen. Mit von der Partie ist auch Shaggys Hund Scooby-Doo, der, genau wie sein Herrchen, immer einer gehaltvollen Zwischenmahlzeit zugetan ist. Wild entschlossen, das Geheimnis zu lüften und die Suspendierung aufheben zu lassen, machen sich die fünf Freunde daran, ihren ersten Fall zu lösen.
