Was ist der Sinn des Lebens? Diese äußerst tiefgründige und philosophische Frage stellt sich der einfache Dorfbewohner und notorische Faulenzer Zed (Jack Black), nachdem er vom Baum der Weisheit gegessen hat. Seine neuerworbene Erleuchtung bringt ihn prompt in so große Schwierigkeiten mit dem Rest seines Dorfes, dass er fliehen muss. Gemeinsam mit seinem Kumpel Oh (Michael Cera), der sich Zed widerwillig anschließt, brechen die beiden Freunde auf zu einer abenteuerlichen Reise quer durch den Anbeginn der Zeitrechnung. Auf der Suche nach einer Antwort geraten sie - immer im denkbar ungünstigsten Moment - in allerlei haarsträubende Situationen, die ihre wildesten Träume übertreffen, und verursachen dabei Katastrophen von wahrhaft biblischen Ausmaßen.
