Nel precedente film si traccia l’origine della centenaria faida tra gli aristocratici vampiri conosciuti come Agenti di Morte ed i loro antichi schiavi, i Lycans. Negli Anni Oscuri, un giovane Lycan chiamato Lucian (Sheen) diviene un potente leader e raduna i lupi mannari per rivoltarsi contro Viktor (Nighy), il crudele re dei vampiri che li ha costretti in schiavitù. Lucian è appoggiato dal suo amore segreto, Sonja (Mitra), nella battaglia contro l’esercito degli Agenti di Morte e nella lotta per la libertà dei Lycans.
