Peter Bretter (Jason Segel) hat sechs Jahre damit verbracht, seine Freundin Sarah Marshall (Kristen Bell) anzuhimmeln und ein unauffälliges Leben im Schatten ihrer Schönheit und ihres Ruhms als Fernsehstar zu führen. Umso härter ist der Aufprall, als sie sich von ihm trennt. Weder mexikanisches Bier noch billiger Lambrusco können ihn aus seinem verheulten Elend retten, und noch schlimmer wird es, als er aus den Nachrichten erfährt, dass sein Nachfolger an Sarah Marshalls Seite der berüchtigte Schürzenjäger und erbärmlich hippe Brit-Rocker Aldous Snow (Russell Brand) ist.


Als selbst ein erfolgreicher One-Night-Stand in Tränen und Beziehungstrauer endet, entscheidet sich Peter für eine emotionale Auszeit - für einen Besinnungsurlaub auf Hawaii.
