Der Ausflug von Löwe Alex (Stimme: Jan Josef Liefers), Zebra Marty (Rick Kavanian), Giraffe Melman (Bastian Pastewka) und Nilpferd Gloria (Claudia Urbschat-Mingues) nach Madagaskar hat den New Yorker Zootieren zwar viel Freude bereitet, doch das Heimweh nach der gewohnten Umgebung ist grer als die Lust auf noch mehr Abenteuer in der freien Wildbahn. Mit Hilfe der pfiffigen Pinguin-Truppe (Die Fantastischen Vier) machen sie ein altes Flugzeugwrack wieder flott und starten mit dem Ziel Wahlheimat Central Park. Doch die Zuverlässigkeit von Air Pinguin ist nicht die beste. Mit dem Lemurenkönig Julien (Stefan Gossler) und seinem Privatsekretär Maurice (Roland Hemmo) an Bord legt die Reisegruppe mitten in der Steppe Afrikas eine Bruchlandung hin. Und wie es der Zufall will, stranden die Zootiere im Reservat von Löwenkönig Zuba (Engelbert von Nordhausen), der seinen im Kindesalter verschollenen Sohn Alex für tot hielt. Umso grer ist die Überraschung, als dieser plötzlich auf die Matte steht und sich als König von New York vorstellt. Doch Konkurrent Makunga (Klaus-Dieter Klebsch) wittert seine Chance, Zuba mit Hilfe einer Intrige, in deren Mittelpunkt der unbedarfte Alex steht, als Alphatier abzulösen
