Der kalifornische Reiseführer-Autor Mike Enslin (John Cusack) schreibt gerne über übernatürliche Phänomene - und hält sie alle für Ammenmärchen. Bei seinen Recherchen stößt er auf ein New Yorker Hotelzimmer, in dem es tatsächlich spuken soll. Ungerührt von den Warnungen des Hotelmanagers Gerald Olin (Samuel L. Jackson) verbringt er die Nacht in Zimmer 1408, in dem seit 1912 über 50 Gäste binnen Stundenfrist den Tod fanden.
