Samen met haar drie kleine poesjes Marie, Toulouse en Berlioz leeft poes Duches in een grote villa bij de lieve, bejaarde en wat excentrieke Madame Bonnefamille. Als de jaloerse en hebberige Butler Edgar toevallig hoort dat Madame na haar overlijden al haar bezittingen aan poes Duchess zal vermaken, weet hij wat hem te doen staat. Hij besluit de “kattige” erfgenamen te katnappen en ze uit de weg te ruimen. Het plan mislukt en op weg naar huis ontmoet ze al snel de avontuurlijke straatkat Thomas O’Malley. Het is aan hem en zijn jazzband van swingende katten om deze slag te winnen. Een nieuw Disney liefdeskoppel is geboren.
